Formulation of medium : 42

MEDIUM 42 - for Lactobacillus pontis

- Man Rogosa Sharp broth 55 g (^Bio-Rad 64234)
- Ultra pure water make up to 1000 ml (^Millipore MilliQ)
- Lactobacillus pontis solution - M0209 18 ml

Add all products except M0209 solution Adjust pH to 6.5 + 0.2 Autoclave at 121°C for 15 minutes Cool to 50°C Add aseptically M0209 solution

Lactobacillus pontis solution - M0209 18 ml

- Maltose 15 g (^Merck 5910)
- Hemin solution - M00149 50 ml
- L-Cystein 5 g (^Sigma C7352)
- Ultra pure water make up to 100 ml (^Millipore MilliQ)

Adjust pH to 6.5 + 0.2 Filter sterilize

Hemin solution - M00149 50 ml

- Hemin chloride 0.1 g (^Merck 3741)
- Sodium hydroxyde 30% 0.1 ml (^Fisher S/4950/PB15)
- Triethanolamine 4 ml (^VWR 28743.290)
- Ultra pure water make up to 96 ml (^Millipore MilliQ)

Filter sterilize
