MEDIUM 88 - for Mycoplasma and Acholeplasma

- PPLO broth 17 g (^Difco 255420)
- Yeast extract solution 25 % - M00263 100 mL
- 0.1 % Phenol red - M00387 20 mL
- Ultra pure water make up to 670 mL (^Millipore MilliQ)
- Sterile supplement - M00273 210 mL

Add all components except sterile supplement Adjust pH to 7.6 + 0.2 Autoclave at 121°C for 15 minutes Add aseptically sterile supplement

Yeast extract solution 25 % - M00263 100 mL

- Yeast extract 250 g (^{BD Difco} 212750)
- Ultra pure water make up to 1000 mL (^Millipore MilliQ)

Adjust pH to 7.0 + 0.2 Autoclave at 121°C for 15 minutes.

0.1 % Phenol red - M00387 20 mL

- Sodium hydroxyde 5 mL (^Prolabo 28 252.293)
- Phenol red 500 mg (^Merck 7241)
- Ultra pure water make up to 500 mL (^Millipore MilliQ)

Adjust pH to 7.0 + 0.2 Autoclave at 121°C for 15 minutes.

Sterile supplement - M00273 210 mL

- Glucose 50 % solution - M00274 10 mL
- Horse serum 200 mL (^Gibco 26050-088)
- Ampicillin 1 g (^{Bristol-myers squibb} 4 3244576 000197 00)

Glucose 50 % solution - M00274 10 mL

- Glucose 50 g (^Sigma G8270)
- Ultra pure water make up to 100 mL (^Millipore MilliQ)

Adjust pH to 7.0 + 0.2 Autoclave at 121°C for 15 minutes.
